      目的 探讨膜联蛋白A2(annexin A2, ANXA2)在登革病毒Ⅱ型(dengue virus type Ⅱ, DENV-2)入胞过程中的作用。方法 通过敲低ANXA2和S100钙结合蛋白A10(S100A10)表达以及抗体阻断实验验证ANXA2在DENV-2复制周期中的作用,并进一步通过激光共聚焦显微镜观察、免疫共沉淀分析明确ANXA2与DENV-2 E蛋白的相互作用。结果 DENV-2感染ANXA2和S100A10敲低的细胞中,DENV-2 RNA水平显著降低;ANXA2抗体和DENV-2 E蛋白抗体可有效抑制DENV-2的入胞效率,细胞内病毒RNA水平显著低于对照组;此外,ANXA2在DENV-2感染细胞中由细胞质募集到细胞膜上,与DENV-2 E蛋白有相互作用。结论 ANXA2是DENV-2入胞过程中重要的宿主分子,有望为抗DENV-2研究提供新的靶点。

      Objective To investigate the role of annexin A2 (ANXA2) in the process of dengue virus typeⅡ (DENV-2) entry into cells. Methods The role of ANXA2 in DENV-2 replication cycle was verified through the knock down of ANXA2 and S100 calcium binding protein A10 (S100A10) expression and antibody inhibition assay. Furthermore, laser scanning confocal microscope and co-immunoprecipitation assay were performed to test interaction between ANXA2 and DENV-2 envelope (DENV-2 E) protein. Results The DENV-2 RNA level was significantly decreased in the DENV-2-infected cells when ANXA2 and S100A10 expression were knocked down. ANXA2 antibody and DENV-2 E protein antibody could effectively inhibit the efficiency of DENV-2 entry into cells, and viral RNA level in cells was significantly lower than that in the control group. In addition, ANXA2 was recruited from cytoplasm to cell membrane, and interacted with DENV-2 E protein during DENV-2 infection in cells. Conclusions ANXA2 is an important host factor during the process of DENV-2 entry into cells, and can serve as the novel target for anti-DENV-2 research.
